Sample interview questions
How would you validate a bioinformatics pipeline for ctDNA variant detection before commercial launch?

I would define intended use and performance requirements first, then evaluate analytical sensitivity, specificity, precision, reproducibility, and robustness using representative samples and controls. I would establish traceable acceptance criteria, assess edge cases such as low allele fractions, document all results, and ensure the validation package supports quality and regulatory expectations.

Describe how you would set evidence-based quality-control thresholds for an NGS diagnostic workflow.

I would examine historical and validation data to identify metrics that predict assay performance, including coverage, read quality, contamination, error profiles, and sample-level yield. Thresholds should be justified by their relationship to false-positive and false-negative risk, tested prospectively, monitored after release, and revised through controlled change management when evidence supports it.

How do you balance hands-on technical leadership with managing a team of bioinformaticians and data scientists?

I set clear technical priorities, decision frameworks, and milestones while delegating ownership to team members based on capability and development goals. I stay close to the highest-risk scientific decisions, use regular reviews to remove blockers, and create an environment where team members can independently propose and defend sound solutions.

Give an example of how you would resolve disagreement between bioinformatics, molecular biology, and software engineering teams.

I would bring the groups together around the shared product requirement and make assumptions, evidence, constraints, and patient-impact risks explicit. I would define measurable experiments or analyses to resolve uncertain points, document the decision rationale, and assign accountable owners and timelines for the agreed path.

What statistical approaches are important when optimizing algorithms for cancer-associated variant detection?

Important approaches include probabilistic error modeling, estimation of sensitivity and specificity, classifier calibration, confidence-interval analysis, multiple-testing considerations, and stratified performance evaluation across variant types and allele fractions. The chosen approach should prioritize clinically meaningful error control and be interpretable enough to support validation and ongoing monitoring.
